The best flights from the States to Greece are by Delta, American and United. All three airlines have overnight flights from the East Coast directly to Athens. Cleveland and other cities have very good connections with the above airlines. 

Samos is a Greek island in the Aegean Sea and it is connected with Athens by a short 40 min flight. Therefore, when you arrive in Athens, you will take the next flight to Samos. 

If you want to see Athens and the sites, a day/night stay is recommended. This can be done at the beginning or the end of the trip. It is also possible to extend the trip to other areas of Greece or other European cities.
