There are many smaller inns and hotels in the charming, centuries-old fishing village of Greenport (an east coast get-away destination about 30-45 minutes from the wedding).

Harborfront Inn at Greenport – a 35 room waterfront hotel with a heated outdoor pool.
American Beech – a trendy 18 room boutique hotel in Greenport with a popular restaurant and boutique.
Sound View Greenport – a 65 room modern motel with a private beach, pool, and restaurant.
Greenporter Hotel – a simple and casual hotel in Greenport with a pool.
The Harvest Inn – a spacious Bed & Breakfast in the middle of the North Fork.
The Zen Hotel – a cute, tiny boutique hotel that recently opened in Greenport.

There are plenty of house rentals in the area. If you are traveling as a big group this is a good option!
Getting There
There are many modes of transportation that can be taken to get to Riverhead. If you choose to drive, please keep in mind that Long Island traffic can be BRUTAL. Please allow yourself extra time for travelling to the end of Long Island.
All Aboard!
If you're from the NYC metro area and plan to take the train from Penn Station to Riverhead, use the Long Island Rail Road (LIRR). Riverhead is a station along the Main Line (Greenport Branch) of the LIRR. ~ 10-20 minute Uber to recommended hotels with room blocks.
Ship Shape
If you're New England-based, making a Ferry Crossing with your car might make more sense than driving around the island. Long Island is served by two different year-round ferry lines that cross the Long Island Sound. The Cross Sound Ferry connects New London, CT to Orient Point, NY. This ferry docks ~ 30 minutes from the venue and nearby hotels. There is also the Port Jeff Steamboat Company, which leaves from Bridgeport, CT and docks at Port Jefferson, NY (~ 45 minutes away from venue). If you decide to go by Ferry, please make sure to reserve your car well ahead of time as ferries book up during peak summer months.
On a Jet Plane
LaGuardia Airport (LGA) and John F. Kennedy Airport (JFK) are both ~1 hour 30 minutes to Riverhead. Feel free to rent a car at the airport and take a ride up the Long Island Expressway.
